CAPILLARY SEQUENCING AND FRAGMENT ANALYSIS

ABI 3730xl

Applied Biosystems 3730xl (2 units).  These two 96-capillary array electrophoresis instruments are used for DNA Sanger sequencing of plasmid or PCR products.  They are also used for fragment analysis of 96-well plates of fluorescently-labelled PCR products.

NEXT GENERATION SEQUENCING

Illumina HiSeq 2000 and 2500

Illumina HiSeq 2500 (2 instruments).  This instrument can run two 8-channel flowcells in high output mode or run two 2-channel flowcells in "rapid run mode."  Currently, maximun read lengths are 100 bp/read.  Depending on the application, run times in rapid run mode range from 10 hours to 2 days.  In rapid run mode, maximum number of reads per run are about 300 M per flowcell and maximum base yield is about 50 GB per flowcell.  In rapid run mode, the instrument collects data quickly and may yield longer read lengths.

Illumina MiSeq

Illumina MiSeq (2 units).  This instrument can rapidly run a single sample or pool of samples.  Currently, maximun read lengths are 300 bp/read.  Depending on the application, run times range from 1 to 2 days.  Maximum number of reads per run are about 35 M per flowcell and maximum base yield is about 8 GB per flowcell.

Illumina NextSeq500 (2 units).  This instrument can rapidly run a single sample or pool of samples.  Currently, maximun read lengths are 150 bp/read.  Depending on the application, run times range from 1 to 2 days.  Maximum number of reads per run are about 800 M per flowcell and maximum base yield is about 120 GB per flowcell.

MICROARRAYS

Illumina BeadStation 500GX

Illumina BeadStation 500GX (includes an Illumina BeadArray Reader, hybridization oven, and orbiting plate shaker).  This microarray system can scan Illumina BeadChips using the Illumina GoldenGate or Infinium chemistries.

Agilent scanner

Agilent G2565CA (microarray scanner with autoloader).  This laser scanning instrument can be used to scan Agilent, Nimblegen, and custom microarrays.  It is capable of scanning slides with 2 micron resolution.

REAL-TIME PCR

ABI Vaii7

Life Technologies / ABI Viia7.  This instrument can be used in 96-well or 384-well format, and can be used for real-time PCR analysis using fluourescent primer/probes or Sybr green.  It can be operated in real-time PCR mode or endpoint analysis mode.  It is also capable of performing melt curve analysis of PCR products.  The facilty's on-line scheduling system is used to schedule time on this instrument.
 
ACCESSORY INSTRUMENTATION

Nanodrop1000 spectrophotometer.  Used for quantification of nucleic acids and uses 1-2 microliters for each measurement.

Invitrogen Qubit spectrofluorometer.  Can quantify DNA/RNA using picogreen flourescence.

Turner Systems spectrofluorometer.  Used for quantifying DNA using fluorescence.

AATI Fragment Analyzer.  Used for quality assessment/quality control (QA/QC) of nucleic acids, typically for assessing RNA integrity and for sizing of DNA/RNA molecules.

Covaris S2 Adaptive Focused Acoustic Disruptor.  Used to randomly-fragment DNA molecules or chromatin by sonication.

Sage Sciences Blue Pippen.  Uses disposable agarose gel cassettes to automatically size select DNA molecules.

Qiagen TissueLyser

Beckman Biomek FX.  Used to perform automated liquid handling protocols using disposable tips.

Beckman Biomek NX.  Used to perform automated liquid handling protocols using fixed tip protocols.

Note on accessory instrumentation:  These instruments are operated by the Genomics Facility staff, but may be made available for use by investigators for their sample preparation, quality control, or sample quantification protocols.  For pricing information and for access and use by investigators, consultation with the Director of the Genomics Facility is required both to coordinate training on the instruments and to schedule instrument usage.  Please contact us.
