Illumina library: small projects
We prepare the libraries according to standard protocols and using commercial kits that yield nearly 100% success rates. All steps are handled manually. In addition, we re-do preparations for samples that fail on the first attempt.
Prep kits for DNA-Seq
From purified gDNA
- Nextera Flex (Illumina) — Faster turnover. Very flexible, can be used for sequencing large and complex genomes, microbiome, and metagenomic samples.
- TruSeq DNA PCR-free (Illumina) — This kit offers more coverage and a higher quality than Nextera Flex.
- TruSeq Nano (Illumina) — This kit is used when there are low amounts of starting DNA. Requires PCR amplification post-library construction.
Prep kits for RNA-Seq
From purified total RNA or purified polyA-selected mRNA
- TruSeq (Illumina) — 3' RNA-seq.
Prep kits for ChIP-Seq
From post-ChIP DNA
- We do not perform the immunoprecipitation step. The DNA library is created with the commercial TruSeq (Illumina) or TruSeq-equivalent (home-made mix that uses the same ligase as the commercial Illumina kit and gives comparable results).
