AVITI24 Sequencing (Genomics Facility)

These sequencing runs are sent out to external providers but supported by the Genomics Facility (including local ordering/sample drop-off, QC checks and data management/distribution/archive), and without the need to acquire a quote/PO outside Cornell.

General Information

  • Why would you sequence your project on the AVITI24:
    • This sequencer is compatible with single barcode libraries as the rolling circle amplification largely prevents index hopping.
    • Supports custom sequencing primers; Read 1, Read 2, Index1 and Index2 (fees may apply).
    • Supports single-end or paired end reads of custom length. Contact us before placing an order to discuss options.
  • Sequencing chemistry: The Element Biosciences AVITI24 is a mid-output benchtop sequencer built on Avidite Base Chemistry (ABC) and rolling circle amplification that circularizes, copies, and rolls each DNA strand into tightly bound polonies, then uses 4-color fluorescent dye-labelled polymers to create ultrastable complexes for highly accurate base detection (Q30 > 90%).
  • Sequencing Output: The system offers a dual flowcell layout that supports up to 1.0 billion reads passing filter per flowcell (2x150bp only). The AVITI24 also supports read lengths of up to 2 x 300 bp. The dual lanes are individually addressable which allows seperate pools to be sequenced on each lane, additional fees would apply. Please contact us if you are interested as our current ordering does not support this option.
  • Illumina Library Compatibility: The AVITI24 is compatible with most Illumina-style adapters and libraries without requiring any conversion, with a few notable exceptions. Before submitting, please confirm your libraries compatibility on the Element Biosciences website. If you are still unsure, please contact us!

         There are unique cases where some libraries (off the shelf and/or custom generated libraries) may have truncations/missing bases which would require a simple end-polish PCR (EP-PCR). This is a simple 4 cycle PCR using the KAPA HiFi Amplification Mix, followed by a bead-based cleanup performed on your library pool – not individual samples. Please contact us before placing an order to discuss if this applies to your project.

  • Quality checks: We will assess the concentration and size distribution of your Illumina samples and will ask you to approve the results before we continue with the sequencing run. If we observe any problems with sample quality (such as excess primers or adapter dimer peaks), we may recommend additional cleanup steps before sequencing (fees may apply). Samples that do not pass the quality checks will be charged for QC only if sequencing is cancelled.
