Protein-protein interaction & protein complex analysis

This analysis interrogates either which proteins interact with which ones, or how many and which proteins interacte in a protein complex. The Proteomics and Metabolomics Facility offers two different workflows with different technologies to do these analyses:

  1.  DSSO-based cross-linking mass spectrometry (XL-MS) for protein-protein interactions and protein complex to determine the identities and spatial orientations of interacting proteins simultaneously by freezing transient interactions through the formation of covalent bonds.
  2. Immuno-precipitation pulldown to specific lure protein and its protein interacting complex, followed-up by either in-gel or in-solution protein identification to figure out which proteins are uniquely present under treated conditions over controls.

Analyses of protein-protein complex can quantitatively compare two groups (e.g. wild type and mutant), and explore what protein interactions have changed between the two groups.
